Response Surface Methodology Based Development and Quantification of Celecoxib and Amlodipine Using RP-HPLC.

A novel RP-HPLC method for estimation of Amlodipine Besylate and Celecoxib using an experimental design approach employing response surface technique was developed validated a C18 column its application in marketed formulation. multivariate optimization conditions carried out organic content the mobile phase, pH, flow rate phase as three independent variables. The aim study to apply methodology effect variables on separation both drugs by faced central composite (FCC) which is this area research. Optimization retention time last eluting peak symmetry performed Derringer’s desirability function potassium dihydrogen phosphate buffer with pH 4. 45, 25 mM, 0.5% Triethylamine acetonitrile isocratic proportion 70:30% v/v 1.2 ml/min found be predicted optimum condition. DoE based development led efficient lower eluted peaks below 7 min indicating novelty linear observed over concentration range 40-240 μg/mL 1-6 μg/ mL Besylate. Limit detection (LOD) limit quantitation (LOQ) were 0.02 0.08μg/mL, 0.0053 0.0162 μg/mL, respectively. successfully accordance ICH guideline acceptance criteria linearity, accuracy, precision, specificity, robustness.
